I was wondering if this process was possible, and viable... Guess this answers it. Thanks for the video. Just a question though, how many times have you gone through this process, and what were your level of results? Has it always worked, or have you had times where it failed?
There are a lot of factors that will effect how long yeast will survive such as the health of the yeast when it was bottled, the strain of yeast being used, how the bottles have been stored etc. A laboratory recovered yeast from a bottle of beer found on a ship that was sunk over 150 years ago so if you have a bottle with some yeast in that you want to use there's never any harm in trying. The method I have described in the video is intended to give you the best chance of success. Good luck.Are you suggesting all bacteria, molds and spores that can be cultured in wort will be killed in 10 seconds ? Do you have a source for that claim ? Or are you suggesting that all beer spoiling microbes will be killed, bearing in mind we aren't dealing with beer at this stage.FYI, all detrimental microbes will be killed within 10 seconds of a rolling boil. No need to boil for 15-20 minutes. Otherwise, informative video.
Experience suggests that it is dependent on yeast strain. Temperature and the health of the yeast at the time it is put into the fridge will also have a bearing. I would suggest using it with in the week considering that yeast cells continually die off and that the aim is to pitch a known number of cells (determined by the starter size). Given that yeast will still strive to populate the wort it is pitched into you may still have success beyond that but the results may be less than optimum.
Using heat is a more thorough method for killing bacteria than using chemical sanitizers. When culturing small quantities of yeast which may be in less than perfect condition, it is important to try to reduce the bacterial load as much as possible or preferably eliminate it altogether in order to ensure that you end up making a yeast starter and not a bacteria starter.
nice job., but you could have saved a lot of time and a fair few steps if you put a filtered breather hole on the litle jar and pressure cooked it for 15 Min's. once done and cool pour you yeast as you did in the video. other than that this helped me a lot with the concepts involved. many thanks.

wow didnt expect to get hostility. i was trying to help you there mate. if you dont have a pc then you can steralise a liquid medium with non pressurised steam and at a push in the microwave. the key is that you would be exposing the contents to the air much less since it will be a sealed filter container i must admit i was impressed at your sterile procedure considering you were holding a camera while you worked. ive seen some YouTube instruction videos that were a right mess.

Don't be so sensitive, no hostility intended. It's just 'one' method of culturing yeast from a bottle. At the time it was made it was a little different from most YT videos and may suit some more than others. What's important is culturing very small quantities of yeast requires a slightly different technique to regular starters and a higher level of sanitation. Another alternative to using a PC is tindalization (see Wikipedia) but as with microwaves and steam there are various drawbacks.

It was especially clever how you transferred the hot wort to the hot glass. Pyrex jugs in the UK are made from Borosilicate glass, the same as laboratory glassware (source wikipedia) which is better for use directly over a flame. Soda Lime isn't unfortunately and is more likely to crack and break. If you can get hold of lab glassware such as a small Erlenmeyer flask that would be even better. You can grow the yeast on in it if you want, just cover the opening with foil.please see the wikipedia article for "Borosilicate glass" and "Erlenmeyer flask" to hopefully make things clearer. Sadly You Tube wont let met post links.
*UPDATE: after exchanging e-mails with the head brewer at Worthingtons, i've since found out that the bottling yeast in White Shield isn't the primary strain (despite what is implied on the back of the bottle). It's still a yeast worth using though if you are prepared to put in the extra work as it required lots of rousing..

This is excellent, thanks Chris! How do you know which ales have viable yeast in their bottles? Very few bottles seem to state "bottle conditioned" these days.
You can only really check by looking for the sediment (or phoning the brewery) but not all bottled conditioned beers use the primary strain,some use a seperate strain for bottling which isn't worth reusing. I only know of 3 good sources for sure, Hop Back, Worthingtons, Coopers and possibly Youngs. Your own bottle conditioned beers are also a good source for yeast and a hassel free way of storing expensive liquid strains. (I'd always buy fresh when using a dried strain). Good luck.
Always store yeast in the fridge and bring it out at least an hour before you intend to use it. Some yeast strains keep better than others, try and get this into a 1-2L starter with a week as such as small quantity is still quite vulnerable. Having said that, i've been experimenting with the yeast in the video (Worthington White Sheild) seems to be bullet proof. Good luck.
